After 48 Hours, Mur...。サイクルロードレースに青春を燃やす高校生たちの成長や友情を描いた人気アニメ「弱虫ペダル」の完全新作劇場版。原作者の渡辺航が原案を担当した、オリジナルストーリーが描かれる。全国大会（インターハイ）で激闘の末に総合優勝を勝ち取った総北高校自転車競技部の小野田坂道は、卒業を控える3年生の指導の下、「全員で支え合い走りぬく」というチーム総北の魂を継承するため、日々の練習に励んでいた。そんなある日、チーム総北に、インターハイの成績優秀チームが出場する「熊本火の国やまなみレース」からの出場招待が届く。打倒総北に燃えるライバルチームや地元・熊本台一など、全国の強豪が集まるレースに向け、坂道たちは士気を高めていくのだが……。。
